Lyophilization is a drying process in which a solvent, usually water, is removed from a frozen material by sublimation under reduced pressure. The material is first solidified, then placed under vacuum so that ice transitions directly to vapor without a bulk liquid phase. This approach suits heat-sensitive substances that would degrade during conventional evaporation. Primary drying removes unbound ice, while secondary drying reduces water that remains adsorbed to the solid matrix. The result is a porous, lightweight solid that can be reconstituted later.
A typical cycle begins with freezing, sometimes including an annealing step to control ice crystal size. Freezing conditions influence the pore network that later allows vapor escape. During primary drying, shelf temperature and chamber pressure are set so heat enters the product while its temperature stays below the collapse or eutectic point. Secondary drying then raises the shelf temperature to desorb bound water and lower residual moisture. Cycle design depends on formulation, fill volume, container type, and equipment capability.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.

The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
Lyophilization removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts liquid water into ice and concentrates dissolved solids. Primary drying then lowers chamber pressure so ice changes directly into vapor without passing through a liquid phase. Secondary drying raises the shelf temperature to remove bound water that remains after ice sublimation. The result is a dry, porous structure that can be reconstituted later.
Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
Quality control for lyophilized materials includes visual inspection of the cake, measurement of residual moisture, and tests for reconstitution time. An acceptable cake is typically uniform and may be slightly porous; shrinkage, meltback, or cracks can indicate process deviations. Analytical methods such as Karl Fischer titration, thermogravimetric analysis, and near-infrared spectroscopy quantify water content. Reconstitution time is recorded because a very slow or incomplete dissolution can signal collapse or aggregation. Stability studies compare samples stored under defined temperature and humidity conditions over months or years.
Regulatory expectations for lyophilized products focus on consistent manufacture and documented stability. Batches are often monitored for moisture, appearance, potency, and sterility where applicable. Process parameters such as shelf temperature, chamber pressure, and drying time are recorded and controlled within validated ranges. Open questions remain about how best to predict long-term stability from short accelerated studies, especially for complex biologics. Variations in freezing rate and ice crystal size can produce differences that are not always visible but may affect performance.
Founded in 1949 by Sally Everett, the chain began its expansion under Sally's son Donald Everett Sr. in 1966, and started franchising restaurants in 1979. As of November 2020, there are 85 Runza restaurants operating: 80 in Nebraska, two in Iowa, two in Colorado, and one in Kansas. The restaurant chain is still owned by the Everett family, and Sally's grandson Donald Everett Jr. serves as President. In addition to the namesake sandwich, the chain serves chili and cinnamon rolls (another Midwest dish), as well as other fast food staples like hamburgers, french fries and onion rings. The chain attempted to expand beyond the region in 1989. Executives tried to open a restaurant in the Latvian republic of the Soviet Union, going as far as shipping two hundred frozen Runza sandwiches to the Soviet Ministry of Agriculture as a part of its negotiations. The deal fell apart after Latvia was invaded by the Soviet government in an attempt to keep it in the Union. Stores did open in the Las Vegas Strip at the Fashion Show Mall's food court and a mall food court in Moline, Illinois but both failed to gain traction and closed within a few years.

== Available forms == DBNPA’s available form is dependent on its intended industrial use. Its pure state is a crystalline solid, with a melting point of 123-126 °C. It is slightly soluble in water (1.5 g per 100 g at 25 °C) but dissolves readily in certain organic solvents like acetone and ethanol. For practical applications, DBPNA is most commonly used in the form of a mixture of 20% active solution combined with water and polyethylene glycol, which enhances its stability and handling in aqueous systems. Its solid forms are also available but are mainly used for packaging and storage; it is often packaged in containers within 25 kg woven bags in order to prevent moisture exposure in storage mechanisms.
It is possible that it may be related to Cirroteuthis, and in that case the two posterior stumps, looking like arms, may be the remains of the lateral fins, for they seem too far back for the arms, unless pulled out of position. On the other hand, they seem to be too far forward for fins. So that they are probably arms twisted out of their true position. However, having examined samples of the mass sent to him by Webb, Verrill concluded that "the creature cannot be an Octopus, but is of cetacean nature." He suggested that "the whole mass represents the upper part of the head of [a sperm whale], detached from the skull and jaw." Webb decided the carcass should be moved further inland so that it would not be lost to the sea forever. With the help of "six horses and strong tackle", it was moved several miles closer to St. Augustine, "to the terminus of a railroad," where it was protected from the tide and drifting sand. Its final resting place was South Beach, Anastasia Island, near the hotel of Dr. George Grant. The St. Augustine carcass became somewhat of a tourist attraction and was visited by large numbers of people. It is unknown what happened to the carcass afterwards. Photographs of the St. Augustine carcass were for a long time thought to be lost, and drawings remained the only pictorial evidence of the event. They were finally rediscovered in 1993 by Marjorie Blakoner of California, who recognised them in an old album. Van Lockwood, one of the original photographers of the St. Augustine carcass, kept an album of photos he had taken between 1885 and 1899.

This effect is called herd immunity. Polio, which is transmitted only among humans, is targeted by an extensive eradication campaign that has seen endemic polio restricted to only parts of three countries (Afghanistan, Nigeria, and Pakistan). However, the difficulty of reaching all children, cultural misunderstandings, and disinformation have caused the anticipated eradication date to be missed several times. Vaccines also help prevent the development of antibiotic resistance. For example, by greatly reducing the incidence of pneumonia caused by Streptococcus pneumoniae, vaccine programs have greatly reduced the prevalence of infections resistant to penicillin or other first-line antibiotics.
Another border crossing to the south may have been opened earlier. An account by Heinz Schäfer indicates that he also acted independently and ordered the opening of the gate at Waltersdorf-Rudow a couple of hours earlier. This may explain reports of East Berliners appearing in West Berlin earlier than the opening of the Bornholmer Straße border crossing.
=== Colette Green === Dr. Colette Green (voiced by Lani Minella) is a Black Mesa scientist and one half of the protagonist team in Half-Life: Decay. In Decay, Dr. Green's role in the experiment is to make preparations in a room below the test chamber and initiate the Anti-Mass Spectrometer to run at 105%. Dr. Gina Cross also enters the same room to fix a jam in the specimen delivery system's lift mechanism, meaning they are both in the same place when the Resonance Cascade finally occurs. Following the disaster, the two team up to fight their way through the facility for survival. They escort Dr. Rosenberg to the surface to call the military for help and then, with the help of Dr. Richard Keller, manage to start a resonance reversal to prevent the dimensional rift from becoming too large to be repaired. The outcome for Dr. Green, along with the rest of the survivors in Decay, is unknown to the other Black Mesa survivors.
δDs of annual precipitation in different regions of the world are shown on the map. The precipitation is more D-enriched near the equator in the Tropics. The δDs generally fall in the range of around −30 ~ −150‰ in the northern hemisphere and −30~+30‰ over land areas of the southern hemisphere. In North America, the δD of average monthly precipitation across regions is lower in January (ranging up to around −300‰ in Canada) than in July (up to around −190‰). The overall mean precipitation is determined by the balance between evaporation of water from the oceans and other surface water and condensation of water vapor in the form of rain. Net evaporation should equal net precipitation, and the δD for precipitation is around −22‰ (global average). The Global Network of Isotopes in Precipitation (GNIP) investigates and monitors the isotopic composition of precipitation at various sites all over the world. The mean precipitation can be estimated by the equation, δ2H = 8.17(±0.07) δ18O + 11.27(±0.65)‰ VSMOW. (Rozanski et al., 1993) This equation is the slightly modified version from the general global meteoric water line (GMWL) equation, δ2H = 8.13δ18O + 10.8, which provides the average relationship between δ2H and δ18O of natural terrestrial waters.
Meanwhile, General Armada, another of the conspirators, tried to get the king to authorize him to appear on his behalf in the Congress of Deputies, but Juan Carlos I refused. In spite of this, Armada went to the Congress where he met with Tejero, to whom he explained his plan to form a concentration government presided by him and asked him to let him address the deputies. Tejero flatly refused because he wanted a purely military government. At one o'clock in the morning, the king, dressed as Captain General as supreme chief of the Armed Forces, addressed the country condemning the military coup and defending the democratic system. It was "the decisive moment to defeat the coup". Two hours later, Milans del Bosch ordered the withdrawal of his troops and the next morning Tejero surrendered, releasing the government and the deputies. The coup of "23-F" had failed. Shortly after, demonstrations in support of the Constitution and in defense of democracy were called, which were the largest of those held up to that time.

=== Sources of veterinarian stem cells === Veterinary applications of stem cell therapy as a means of tissue regeneration have been largely shaped by research that began with the use of adult-derived mesenchymal stem cells to treat animals with injuries or defects affecting bone, cartilage, ligaments and/or tendons. There are two main categories of stem cells used for treatments: allogeneic stem cells derived from a genetically different donor within the same species, and autologous mesenchymal stem cells, derived from the patient before use in various treatments. A third category, xenogenic stem cells, or stem cells derived from different species, are used primarily for research purposes, especially for human treatments.

Another independent approach is to use oligoadenylated anti-telomerase antisense oligonucleotides and ribozymes to target telomerase RNA, leading to the dissociation of the RNA and to apoptosis (Figure 5). The fast induction of apoptosis through antisense binding may be a good alternative to the slower telomere shortening.
=== Tumor detection === In cancer research, colloidal gold can be used to target tumors and provide detection using SERS (surface enhanced Raman spectroscopy) in vivo. These gold nanoparticles are surrounded with Raman reporters, which provide light emission that is over 200 times brighter than quantum dots. It was found that the Raman reporters were stabilized when the nanoparticles were encapsulated with a thiol-modified polyethylene glycol coat. This allows for compatibility and circulation in vivo. To specifically target tumor cells, the polyethylenegylated gold particles are conjugated with an antibody (or an antibody fragment such as scFv), against, e.g. epidermal growth factor receptor, which is sometimes overexpressed in cells of certain cancer types. Using SERS, these pegylated gold nanoparticles can then detect the location of the tumor. Gold nanoparticles accumulate in tumors, due to the leakiness of tumor vasculature, and can be used as contrast agents for enhanced imaging in a time-resolved optical tomography system using short-pulse lasers for skin cancer detection in mouse model. It is found that intravenously administered spherical gold nanoparticles broadened the temporal profile of reflected optical signals and enhanced the contrast between surrounding normal tissue and tumors.

Freezing only converts liquid to solid. Lyophilization adds vacuum and controlled warming so frozen solvent sublimes, leaving a dry porous solid. The two steps are related but not interchangeable.
Reduced pressure keeps the solvent below its triple point, allowing ice to become vapor without melting. Vacuum also helps remove water vapor from the product chamber. The exact pressure is chosen with the formulation and equipment.
Residual moisture is water that remains in the dried solid after secondary drying. It is often measured by Karl Fischer titration, near-infrared spectroscopy, or thermogravimetry. Acceptable levels depend on the material and its stability profile.
Residual moisture can influence chemical degradation, cake collapse, and long-term stability. Low moisture levels usually improve stability, but each product has an optimal range.
