Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
Quality assessment of a lyophilized product includes cake appearance, residual moisture, reconstitution time, and container closure integrity. A uniform, porous cake suggests that freezing and drying stayed within the formulation's design space. Cracks, shrinkage, meltback, or a glassy film can indicate thermal abuse or a formulation problem. Analysts also test for subvisible particles and sterility when the product requires those specifications. Visual inspection alone cannot confirm biological activity or chemical stability, so it is combined with analytical methods.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
| Property | Value | Notes |
|---|---|---|
| Cake appearance | Uniform porous plug | Cracks, shrinkage, or meltback suggest process deviation. |
| Reconstitution time | 10 seconds to 5 minutes | Depends on cake structure, diluent, and agitation. |
| Typical storage humidity | Below 60% relative humidity | Lower humidity limits moisture uptake by hygroscopic cakes. |
| Container closure | Glass vial, elastomer stopper, crimp seal | Seal integrity limits moisture and oxygen ingress. |
| Common moisture test | Karl Fischer titration | Measures residual water content in the dried solid. |
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
After lyophilization, a product's quality depends on residual moisture, cake appearance, and reconstitution time. Residual moisture is often measured by Karl Fischer titration or thermogravimetric analysis. A low moisture content can slow chemical degradation, but overly dry cakes may be brittle or slow to dissolve. Stability studies track these attributes over months under defined temperature and humidity conditions. Batch records link these measurements to specific process runs and help identify trends before a product fails specification.
Storage conditions for dried products usually aim to exclude moisture and oxygen. Vials are sealed under vacuum or with an inert gas, and stoppers must maintain a barrier during transport. Temperature recommendations vary; some materials remain stable at room temperature, while others need refrigeration or frozen storage. Humidity control is critical because dried cakes can absorb water rapidly once a container is opened. Desiccant packs and moisture-barrier bags add further protection during shipping.
Quality control also examines cake structure, color, and reconstitution behavior. A collapsed or shrunken cake can indicate a thermal excursion during drying. Analytical methods such as X-ray diffraction, differential scanning calorimetry, and near-infrared spectroscopy can detect crystallinity or moisture distribution. Regulatory expectations focus on validated assays and lot-to-lot consistency. Questions remain about how well accelerated stability tests predict long-term behavior for every formulation. Visual inspection remains common but is subjective without trained reviewers and reference images.
==== Pharmacokinetics ==== Due to high lipid solubility, cannabidiol is poorly absorbed in the intestine. The absorbed cannabidiol accumulates in adipose tissue or albumin (proteins in blood), which prolongs its elimination from the body. Most absorbed cannabidiol is converted into other metabolites by various enzymes in the liver and intestine, including CYP2C19 and CYP3A4. Cannabidiol is mainly excreted from the body with faeces.
== Presence == In cephalochordates (lancelets), the notochord persists throughout life as the main structural support of the body. In tunicates, the notochord is present only in the larval stage, becoming completely absent in the adult animal, and the notochord is not vacuolated. In all vertebrates other than the hagfish, the notochord is present only during early embryonic development and is later replaced by the bony and/or cartilaginous vertebral column, with its original structure being integrated into the intervertebral discs as the nucleus pulposus.
== Notes == ^α Three countries - Mauritius, Saint Lucia, and Saudi Arabia - have not had figures reported by the FAO since the 1990s but are still included in the data; all three continue to produce coffee at a small scale. They are counted here as producing nations but not included in the global totals. ^β Values for China are estimated by the ICO.

Illicit drug trafficking in the West Indian Ocean (WIO) has increasingly become a major security concern for many States in the region, and is gaining international attention. The Indian Ocean borders 24 states, and accounts for a third of the world’s ocean area. Until recently, other challenges, such as piracy off the coast of Somalia, has been at the forefront of international action. However, the utilisation of the Southern route by drug traffickers, and the consequent issues this has caused, has led to increased focus on how to tackle this issue. The two main producers of heroin for the global market are frequently referred to as the Golden Triangle, including Myanmar, Thailand and Laos, and the Golden Crescent, which includes Afghanistan, Pakistan and Iran. Specifically Afghanistan and Pakistan as the two largest heroin and opium producers and exporters via the WIO. The three traditional ways of drug trafficking out of Afghanistan are; the Balkan route through Iran and Turkey via Eastern Europe to western and central European markets; the Northern Route via central Asia to Russia; and the Southern Route through Pakistan and across the Indian Ocean to Eastern African States and onward to consumer markets predominantly in Europe, Asia and the US. The increased use of the southern route via the WIO may be contributed to many factors, including better law enforcement on land-based routes, and the relatively weak enforcement measures concerning trafficking at sea.
The classical description of myiasis is according to the part of the host that is infected. This is the classification used by ICD-10. For example: dermal sub-dermal cutaneous (B87.0) creeping, where larvae burrow through or under the skin furuncular, where a larva remains in one spot, causing a boil-like lesion nasopharyngeal, in the nose, sinuses or pharynx (B87.3) ophthalmic or ocular, in or about the eye (B87.2) auricular, in or about the ear gastric, rectal, or intestinal/enteric for the appropriate part of the digestive system (B87.8) urogenital (B87.8) Another aspect is the relationship between the host and the parasite which provides insight into the biology of the fly species causing the myiasis and its likely effect. Thus the myiasis is described as either: obligatory, where the parasite cannot complete its life cycle without its parasitic phase, which may be specific, semispecific, or opportunistic facultative, incidental, or accidental, where it is not essential to the life cycle of the parasite; perhaps a normally free-living larva accidentally gained entrance to the host Accidental myiasis commonly is enteric, resulting from swallowing eggs or larvae with one's food. The effect is called pseudomyiasis. One traditional cause of pseudomyiasis was the eating of maggots of cheese flies in cheeses such as Stilton. Depending on the species present in the gut, pseudomyiasis may cause significant medical symptoms, but it is likely that most cases pass unnoticed.
In the 1954 version of Casino Royale, Bond was an American agent working with the British to destroy a ruthless Soviet agent in France, but became more widely known as Agent 007, James Bond, of Her Majesty's Secret Service, who was played by Sean Connery until 1971 and by several actors since. Although Bond films often used the Cold War as a backdrop, the Soviet Union itself was almost never Bond's enemy, that role being more often left to fictional and apolitical criminal organisations (like the infamous SPECTRE). However, Red China was in league with Bond's enemies in the films Goldfinger, You Only Live Twice and The Man With the Golden Gun, while some later movies (Octopussy, The Living Daylights) featured a rogue Soviet general as the enemy. TASS Upolnomochen Zayavit... (TASS is Authorized to Announce ... ) – a Soviet TV series based on Julian Semenov's novel. The plot of the movie is set around fictional African country Nagonia, where CIA agents are preparing a military coup, while KGB agent Slavin is trying to prevent it. Slavin succeeds by blackmailing the corrupt American spy John Glebe. The Falcon and the Snowman is a 1985 film directed by John Schlesinger about two young American men, Christopher Boyce and Daulton Lee, who sold United States security secrets to the Soviet Union. The film is based upon the 1979 book The Falcon and the Snowman: A True Story of Friendship and Espionage by Robert Lindsey.
Trump's cabinet choices were described by news media as valuing personal loyalty over relevant experience, and for having a range of conflicting ideologies and "eclectic personalities". It was also described as the wealthiest administration in modern history, with over 13 billionaires chosen to take government posts. He nominated or appointed 23 former Fox News employees to his administration. Notably, Trump's nomination of Scott Bessent as Secretary of the Treasury made Bessent the highest ranking openly LGBTQ person to serve in the United States government. In March 2026, The Wall Street Journal reported that Trump was planning to fire Kristi Noem after a Senate Judiciary Committee hearing during which she was criticised for her actions during her term of office, including her handling of the killings of Renée Good and Alex Pretti, inappropriate relations with her advisor, Corey Lewandowski, and mismanagement of funds. On March 5, Trump announced her reassignment to a new position, "Special Envoy for The Shield of the Americas", and announced Oklahoma senator, Markwayne Mullin as her successor as the secretary of homeland security. Noem is the first Cabinet official to be removed from her post during Trump's second presidency.

Collagenase 3 is an enzyme that in humans is encoded by the MMP13 gene. It is a member of the matrix metalloproteinase (MMP) family. Like most MMPs, it is secreted as an inactive pro-form. MMP-13 has a predicted molecular weight around 54 kDa. It is activated once the pro-domain is cleaved, leaving an active enzyme composed of the catalytic domain and the hemopexin-like domain PDB: 1PEX. Although the actual mechanism has not been described, the hemopexin domain participates in collagen degradation, the catalytic domain alone being particularly inefficient in collagen degradation. During embryonic development, MMP-13 is expressed in the skeleton as required for restructuring the collagen matrix for bone mineralization. In pathological situations it is highly overexpressed; this occurs in human carcinomas, rheumatoid arthritis and osteoarthritis. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The protein encoded by this gene cleaves type II collagen more efficiently than types I and III. It may be involved in articular cartilage turnover and cartilage pathophysiology associated with osteoarthritis. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3.
The middle third of the vagina involves the urogenital diaphragm. It is supported by the levator ani muscles and the lower portion of the cardinal ligaments. The lower third is supported by the perineal body, or the urogenital and pelvic diaphragms. The lower third may also be described as being supported by the perineal body and the pubovaginal part of the levator ani muscle.

Many dried cakes are hygroscopic and can adsorb water during storage or handling. Absorbed moisture may lower the glass transition temperature and promote chemical reactions. Sealed packaging and controlled humidity reduce this risk.
Cake collapse usually means the product became too warm during the drying cycle. The dried matrix loses porosity and may appear shrunken or glassy. Collapse can slow reconstitution and may signal altered stability, though not every collapsed cake fails specifications.
Karl Fischer titration is a common method for measuring residual water in lyophilized solids. Loss on drying and thermogravimetric analysis are also used in some settings. The chosen method should be validated for the specific formulation and moisture range.
Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
