Quality control for lyophilized materials includes visual inspection of the cake, measurement of residual moisture, and tests for reconstitution time. An acceptable cake is typically uniform and may be slightly porous; shrinkage, meltback, or cracks can indicate process deviations. Analytical methods such as Karl Fischer titration, thermogravimetric analysis, and near-infrared spectroscopy quantify water content. Reconstitution time is recorded because a very slow or incomplete dissolution can signal collapse or aggregation. Stability studies compare samples stored under defined temperature and humidity conditions over months or years.
Regulatory expectations for lyophilized products focus on consistent manufacture and documented stability. Batches are often monitored for moisture, appearance, potency, and sterility where applicable. Process parameters such as shelf temperature, chamber pressure, and drying time are recorded and controlled within validated ranges. Open questions remain about how best to predict long-term stability from short accelerated studies, especially for complex biologics. Variations in freezing rate and ice crystal size can produce differences that are not always visible but may affect performance.
The physics of lyophilization couples heat transfer, mass transfer, and phase behavior. Sublimation requires a vapor pressure difference between the ice front and the chamber, and the dried layer adds resistance to vapor flow. Amorphous formulations are characterized by a glass transition temperature of the maximally freeze-concentrated solute, often denoted Tg'. Crystalline bulking agents can provide structure, while amorphous excipients stabilize labile components. Open questions remain about spatial heterogeneity, edge effects, and how laboratory cycles scale to production.
Lyophilization is a drying process in which a solvent, usually water, is removed from a frozen material by sublimation under reduced pressure. The material is first solidified, then placed under vacuum so that ice transitions directly to vapor without a bulk liquid phase. This approach suits heat-sensitive substances that would degrade during conventional evaporation. Primary drying removes unbound ice, while secondary drying reduces water that remains adsorbed to the solid matrix. The result is a porous, lightweight solid that can be reconstituted later.
A typical cycle begins with freezing, sometimes including an annealing step to control ice crystal size. Freezing conditions influence the pore network that later allows vapor escape. During primary drying, shelf temperature and chamber pressure are set so heat enters the product while its temperature stays below the collapse or eutectic point. Secondary drying then raises the shelf temperature to desorb bound water and lower residual moisture. Cycle design depends on formulation, fill volume, container type, and equipment capability.

| Residual moisture specification | 0.5–3.0% w/w | Product-specific; measured after drying |
| Common moisture method | Karl Fischer titration | Coulometric or volumetric; detects water content |
| Cake appearance | Uniform and porous | Collapse, meltback, or cracks are deviations |
| Reconstitution time | Seconds to several minutes | Depends on formulation, cake structure, and diluent |
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Industries use lyophilization for pharmaceuticals, biological products, and food preservation. In the pharmaceutical sector, it extends the shelf life of injectable drugs, vaccines, and proteins that are unstable in aqueous solution. Food manufacturers apply freeze-drying to coffee, fruits, and ready meals to retain flavor and texture. The process is energy-intensive and requires specialized equipment, which limits its use to high-value products. Ongoing research examines how formulation and process parameters affect the quality of the final dried product.
Lyophilization, also known as freeze-drying, is a process that removes water from a material by freezing it and then reducing pressure to allow ice to sublimate directly into vapor. The method begins with a freezing step that solidifies the water content. Next, primary drying lowers the pressure below the triple point of water, enabling sublimation without passing through a liquid phase. A final secondary drying step removes bound water through desorption. This sequence produces a dry, porous cake that can be reconstituted later.
The process relies on the phase diagram of water, where the triple point marks the conditions at which ice, liquid water, and vapor coexist. By maintaining pressure below this point, typically around 0.01 to 0.1 millibar, sublimation becomes the dominant mechanism. Formulations often include excipients such as sugars or polymers that act as lyoprotectants and bulking agents. These additives help preserve the structure of the active ingredient and prevent collapse during drying. The choice of excipient and freezing rate influences the final cake morphology and stability.
=== Intravesical instillation === Sodium hyaluronate can be instilled into the bladder for the treatment of various forms of cystitis and associated bladder pain, by replenishing the glycosaminoglycan layer of the bladder urothelium.
=== Physical activity and transportation === "In 2020, 24.2% of adults aged 18 and over met the 2018 Physical Activity Guidelines for Americans for both aerobic and muscle-strengthening activities." This means that more than 75% of citizens did not get enough physical activity that is necessary for their well-being. With the increase in the usage of automobiles in the 20th century, physical activity has become less necessary. As of 2024, 91.7% of households in the US have at least one vehicle. A researcher from the University of Illinois, Sheldon Jacobson, discusses the surge of passenger vehicle usage in relation to the surge of obesity rates between the 1950s and today: "For the last 60-plus years, we've literally built our society around the automobile and getting from point A to point B as quickly as we can. Because we choose to drive rather than walk or cycle, the result is an inactive, sedentary lifestyle. Not coincidentally, obesity also became a public health issue during this period." Since obesity is affected by not only energy intake but also by energy output, the shift to driving has resulted in less physical activity and a higher risk of obesity.
Gas chromatography-mass spectrometry (GC-MS) is a two-dimensional chromatography technique that combines the separation technique of gas chromatography with the identification technique of mass spectrometry. GC-MS is the single most important analytical tool for the analysis of volatile and semi-volatile organic compounds in complex mixtures. It works by first injecting the sample into the GC inlet where it is vaporized and pushed through a column by a carrier gas, typically helium. The analytes in the sample are separated based upon their interaction with the coating of the column, or the stationary phase, and the carrier gas, or the mobile phase. The compounds eluted from the column are converted into ions via electron impact (EI) or chemical ionization (CI) before traveling through the mass analyzer. The mass analyzer serves to separate the ions on a mass-to-charge basis. Popular choices perform the same function but differ in the way that they accomplish the separation. The analyzers typically used with GC-MS are the time-of-flight mass analyzer and the quadrupole mass analyzer. After leaving the mass analyzer, the analytes reach the detector and produce a signal that is read by a computer and used to create a gas chromatogram and mass spectrum. Sometimes GC-MS utilizes two gas chromatographers in particularly complex samples to obtain considerable separation power and be able to unambiguously assign the specific species to the appropriate peaks in a technique known as GCxGC-(MS).
All serpins share a common structure (or fold), despite their varied functions. All typically have three β-sheets (named A, B and C) and eight or nine α-helices (named hA–hI). The most significant regions to serpin function are the A-sheet and the reactive centre loop (RCL). The A-sheet includes two β-strands that are in a parallel orientation with a region between them called the 'shutter', and upper region called the 'breach'. The RCL forms the initial interaction with the target protease in inhibitory molecules. Structures have been solved showing the RCL either fully exposed or partially inserted into the A-sheet, and serpins are thought to be in dynamic equilibrium between these two states. The RCL also only makes temporary interactions with the rest of the structure, and is therefore highly flexible and exposed to the solvent. The serpin structures that have been determined cover several different conformations, which has been necessary for the understanding of their multiple-step mechanism of action. Structural biology has therefore played a central role in the understanding of serpin function and biology.
From April to May 2024, Rio Grande do Sul was hit by massive flooding, which also severely hit Porto Alegre. This was partly seen as, in addition to the sheer amount of precipitation, insufficient pumping of water, as trash and rubble had blocked many manholes in the city. Melo, during the flooding, affirmed that the city government knew that there was a possibility of strong rains and was not taken by surprise. He denied, however, that the city government had delayed its response and put the responsibility on the new round of floodings for the high volume, as well as the population, blaming them for the trash strewn throughout the city. During the floods, the Guaíba hit its highest ever historical level, leaving various parts of the city submerged and many people stranded. Due to the severity of the situation, the municipality set up 21 centers to receive thousands of people who were forced to leave their homes. The flooding was also marked by conflicts between Melo and the federal government, during which he criticized what he saw was the lack of support from them, despite having also delayed a formal emergency request for days. During this period, federal deputy Fernanda Melchionna and city councilman Roberto Robaina, both from PSOL, sued Melo. They both requested that the State Attorney's Office to start an investigation against Melo due to the fact that the municipal government had not sent the federal government the data of families in Porto Alegre hit by the floods.

The mass spectrometry of proteins has long been a useful technique for identifying posttranslational modifications and, more recently, for probing protein structure. Most proteins are difficult to purify in more than milligram quantities, even using the most modern methods. Hence, early studies focused on proteins that could be purified in large quantities, e.g., those of blood, egg white, various toxins, and digestive/metabolic enzymes obtained from slaughterhouses. Many techniques of protein purification were developed during World War II in a project led by Edwin Joseph Cohn to purify blood proteins to help keep soldiers alive. In the late 1950s, the Armour Hot Dog Co. purified 1 kg (= one million milligrams) of pure bovine pancreatic ribonuclease A and made it available at low cost to scientists around the world. This generous act made RNase A the main protein for basic research for the next few decades, resulting in several Nobel Prizes.
== Catabolism == Degradation of glycosphingolipids occurs in the lysosome, which contains digestive enzymes in animal cells. The lysosome breaks down the glycosphingolipid to its primary components, fatty acids, sphingosine, and saccharide.
In March 2020, it was announced that a secret six-month DEA operation known as "Operation Python" resulted in the arrest of 600 CJNG operatives and the seizure of $20 million in cash. The number of arrests was revised to 750, though it was still thought the CJNG could still partake in trafficking within the U.S. so long as it keeps its base of operations in Mexico. On 2 June 2020, Mexico's Financial Intelligence Unit released a statement revealing that as a result of joint operation with the DEA, the agency was able to locate "a large number of members of said criminal group, as well as its largest financial operators and companies used in money laundering". Mexico's Financial Intelligence Unit, a part of the finance ministry tasked with combating and preventing money laundering, then managed to have 1,770 bank accounts of CJNG-linked individuals frozen. Bank accounts to 16 CJNG-linked companies and two trusts linked to the CJNG were frozen as well. The move came after armed assailants stormed a live CJNG-provided concert on 29 May 2020 in the Veracruz city of Tierra Blanca, injuring two and killing six. Those killed included the regional CJNG leader, and also owner of the local El Sol de Tierra Blanca newspaper, Francisco Navarette Serna and the full lineup of La Calle, the band Serna was performing with. On 3 June, it was reported the amount of CJNG-linked assets which were frozen totaled $1.1 billion.
== Applications == Gold fingerprinting characterizes a gold sample or gold-containing item by analyzing its trace elements, identifying the sample by its mineralizing event and linking it to a specific mine or bullion source. Elements that are measured above detection limits can be used for gold fingerprinting and geochemical characterization. For this technique to be effective in identifying the origins of gold, a database of fingerprinted samples from mines and bullion sources is required. This technique has been used to assert claims over stolen or relocated gold, even in instances where it has been salted—deliberately blended with gold from disparate origins. Beyond this, gold fingerprinting serves as a tool in identifying the often obscure provenance of gold artifacts.
CBD is a 5-HT1A receptor agonist, which is under laboratory research to determine if it has an anxiolytic effect. It is often claimed that sativa strains provide a more stimulating psychoactive high while indica strains are more sedating with a body high. However, this is disputed by researchers. There is currently no consistent evidence that cannabis with higher CBD to THC ratios reduced THC induced psychotic symptoms . A 2015 review found that the use of high CBD-to-THC strains of cannabis showed better cognitive function and both lower risk for developing psychosis, as well as a later age of onset of the illness, compared to cannabis with low CBD-to-THC ratios.

Most are held in sealed containers at controlled temperatures, often 2–8 °C, while some require frozen storage. Protection from moisture and light helps preserve the dry matrix. Exact conditions are set by the manufacturer or study protocol.
Cake collapse suggests the material exceeded its collapse temperature during drying or later absorbed moisture. It can lead to slower reconstitution, uneven moisture, and reduced stability. Appearance alone may not reveal the cause, so process records and moisture tests are used together.
Some residual moisture is common and may be acceptable within a defined range. Very low moisture can alter stability or increase brittleness, while high moisture promotes hydrolysis and microbial risk. Specifications are based on product-specific stability data.
Freezing only converts liquid to solid. Lyophilization adds vacuum and controlled warming so frozen solvent sublimes, leaving a dry porous solid. The two steps are related but not interchangeable.
